A Study on Immunologic changes in Rat Model of Depression

Background/Objective: To investigate the immunologic changes and the effects of aspirin on
behavior and immune factors in rat model of depression.

Method: 30 male adult SD rats, 300+/-50g, devided into three groups by average and random
adaptive raising in animal laboratory for one week, then measured sucorse preference test
(SPT)and forced swimming test (FST). After that one group received chronic unpredictive mild
stimulation(CUMS)for 4 weeks with 2 weeks of aspirin gavage of 50mg/kg; one group received
CUMS for 4 weeks with 2 weeks of NS gavage; and one group was raised in nomal condition with 2
weeks of NS gavage.Then the three groups was tested SPT and FST again. After behaviortests,the
rats were measured the levels of BDNF、CRP、IFN-γ、IL-1β、IL-2、PGE-2、IL-6、IL-10 and TNF-α
in serum and the supernatant of brain homogenate.

Result: After CUMS treatment, the immobility time of FST was significantly increased and the
swimming time and the percentage of sucrose preference were significantly decreased in rat model of
depression compared the control group. There were no any diffences in all cytokines assessments in
serum between the depressive rats and the control rats. However,after aspirin feeding, all cytokines
decreased but BDNF in serum in the aspirin group compared the NS group (P<0.05). Only IL-10
decreased in the supernatant of brain homogenate of depressive rat model compared with the control
(P<0.05). BDNF and IFN-γ decreased in the supernatant of brain homogenate after ouabain
treatment compared with the control, and aspirin gavage can only reverse IFN-γ reduction.

Conclusion: There were some immunologic changes in rat model of depression and included
adaptive and innate immunity. Aspirin may has some effect on the daptive immunity and innate
immunity .
